A Simplified Method for the Cultivation of Endamoeba Histolytica

1. A medium consisting of Locke solution containing one partto seven of inactivated human, horse, or rabbit blood serumhas been found very efficient in the cultivation of Endamoebahistolytica. The best results were obtained when inactivatedhuman blood serum was used.
2. The excellent growth of Endamoebahistolytica obtained inthis medium demonstrates that coagulatedegg or blood agar slantsused as a sub-stratum, are not essentialfor the growth anddevelopment of this species, and that neitheregg or blood areessential as a part of the food supply of thisspecies.
3. In this medium Endamoeba histolytica has remainedalive andmotile for a period of eleven days and successfultransfershave been obtained in the medium after a period ofeight days.The best results in transferring cultures are obtainedby makingtransfers every twenty-four or forty-eight hours.
4. Transfers of Endamoeba histolytica, in this medium, madeat intervals of twenty-four and forty-eight hours, have beencontinued for over three months in this laboratory and the amoebaehave not changed in morphology or in their capability for reproductionduring this time.
